Mesoporous Silica Nanoparticles for pH-Responsive Delivery of Iridium Metallotherapeutics and Treatment of Glioblastoma Multiforme

Using nanoparticles for controlled drug delivery to cancer, in response its weakly acidic environment, represents a promising approach toward increasing the effectiveness and reducing adverse effects of cancer therapy. Hence, aim this study is construct novel mesoporous silica nanoparticle (MSN)-based acidification-responsive systems targeted Herein, surface MSN covalently functionalized with Ir(III)-based complex through pH-cleavable hydrazone-based linker characterized by nitrogen sorption, SEM, FTIR, EDS, TGA, DSC, DLS, zeta potential measurements. Enhanced release Ir(III)-complexes evidenced UV/VIS spectroscopy at environments (pH 5 pH 6) comparison physiological conditions. The vitro toxicity prepared materials tested on healthy MRC-5 cells while their efficient treatment glioblastoma multiforme demonstrated U251 cell line.

Gating of mesoporous silica nanoparticles (MSNs) with the stimuli-responsive poly(β-amino ester) has been achieved. This hybrid nanocarrier releases doxorubicin (DOX) under acidic conditions or in the presence of porcine liver esterase. The DOX loaded poly(β-amino ester)-capped MSNs reduce cell viability when tested on MDA-MB-231 human breast cancer cells.
